FLOW consortium to acquire a complete understanding of the cell's quality control network.

In the 2024 round of NWO Gravitation grants, the new consortium FLOW was awarded 23 million euros. Running from 2024 to 2034, FLOW will investigate the cellular factors that determine the fate of proteins throughout their life and learn how to influence that fate.

A chemically modified version of the antibiotic vancomycin is much more effective against MRSA and other Gram-positive bacteria than the original. Leiden university professor Nathaniel Martin and his group developed this guanidine modified lipoglycopeptide. They recently published their findings in the journal Science Translational Medicine.

A promising target for cancer therapeutics is the protease family of cathepsins. Researchers from the Swiss EPFL institute in collaboration with Dutch researchers from Radboudumc in Nijmegen succeeded in developing a highly specific, targeted way of inhibiting cathepsin activity. The results were published in May 2024 in the journal Nature Chemical Biology.

Postdoctoral project: Innate immune activation and sterile inflammation in microglia in human brain

Timo Oosenbrug is fascinated by immune receptors. "It is the first line of defense against viruses," he explains. "But what makes it even more interesting is what happens when these receptors keep firing all the time, causing sterile inflammation. The translation to this clinical condition is exciting."

Remarkably, we don't know everything about the common cold virus HKU1, part of the coronavirus family. ICI researcher Joost Snijder (Utrecht University) and his team recently not only proved where and how HKU1 binds to the host cell, but also showed how this binding triggers a remarkable conformational change of the spike protein that is fundamental to how this virus enters the cell.

Early 2024, a remarkable publication appeared in Science in which Florian Winau at Harvard and his team describe how they discovered the function of a specific glycolipid called Gb3 found on B cells. From their results, they conclude that Gb3 has the potential to act as an adjuvant to improve vaccines against viral infections such as influenza.

PhD project: Targeting metabolism of the enteric glial cell in Parkinson’s disease

The enteric nervous system possibly plays a key role in Parkinson’s disease, making it a target for possible treatment. Anastasia Markidi at Utrecht University set studies the effects of bacterial metabolites on enteric glial cells.

Enebie Ramos Cáceres is a jack of all trades: his PhD project comprises chemistry and immunology and he is doing all that on his own. He not only develops chemical probes to study key enzymes related to NETosis, relevant to Alzheimer's disease, but he also collects, prepares and studies human blood samples to test his probes.

Omics data sets are an increasingly important part of the research of wet-lab cell biologists and immunologists. But they are usually not equipped with enough bioinformatics or coding skills to be able to analyse these data sets. A first for PhD students in the ICI program, this spring they had the opportunity to participate in the online training "Omic data analysis and visualisation using R" which focuses precisely on these skills.

Targeted anti-cancer therapies are the future of cancer treatment. Immunotherapy is booming, and antibody-drug conjugates (ADC) are a fast growing class of therapeutics. These consist of a cytotoxic drug connected to an antibody that targets a tumor marker. Although much research is aimed at elements of ADCs such as the drug payload or constructing nanobodies, the international research program TACT is bringing all aspects of this research together, from fundamental chemistry to the initial stages of biological assessment. Herpes viruses can invade the nervous system and stay there unnoticed for a long time, before causing disease. But not much is known about the interactions between the virus and the nerve cells. Sabrina Krepel finds out using advanced techniques such as mass spectrometry and electron microscopy.

Microglia play a crucial role in neuronal diseases such as MS or Alzheimer's. But there is a large gap in knowledge about the processes involving microglia and the development of these and other neurological disorders. Without understanding of the fundamentals, there is no chance of a cure. PhD student Viktor al-Naqib at NIN studies these fundamentals in cooperation with chemical immunologists in Leiden.

Thermal peptide exchange technology. The term sounds like a simple lab protocol. But at a closer look it's a perfect example of multidisciplinary, multi-group research where a fundamental background, a good idea and an interest in technology lead to new insights and relevant medical applications. UbiQ was founded in 2010 as a spin-off of the Ovaa-lab at the Netherlands Cancer Institute. The small company develops reagents for ubiquitin research and is involved occasionally in drug discovery projects, says co-founder Farid El Oualid. "Our mission is ultimately to help patients. With UbiQ, we aim to explore all possible drug targets using our technology, and to facilitate others to do their research by providing reagents."

It's an unconventional ICI-project in many ways. LUMC researcher Aysegul Sapmaz wrote the proposal to study ubiquitylation of Rab-proteins back in 2019 together with Huib Ovaa and Sjaak Neefjes. She then moved on to do the research herself in close collaboration with PhD student, Esther ter Linden from the Neefjes group.

A superfamily of four-transmembrane proteins called tetraspanins are organizers of the membranes of immune cells. Although ubiquitous, understanding the precise role of some of these tetraspanins in the complement system is the challenge in this combined project in Utrecht and Leiden.

An imbalance between two types of T cells causes problems in autoimmune diseases and cancer. A selective intervention in the activity of these cells could be a therapeutic strategy, but the question is: how? Researchers in Utrecht and Leiden are finding unique metabolic features that discriminate between the two types.

Bloed- en beenmergkanker zijn goed te behandelen met een stamceltransplantatie van een donor, maar soms keert de tumor terug. Onderzoekers van het Radboudumc onder leiding van Harry Dolstra ontwikkelden een nieuw dendritische celvaccin. Dat heractiveert de afweercellen van de donor waardoor de tumor wordt opgeruimd. Ze toonden aan dat het vaccin veilig is voor patiënten.

Jongens die chemotherapie krijgen tegen kanker zijn later vaak onvruchtbaar. Ans van Pelt ontwikkelde met steun van ZonMW een manier om uit een stukje ingevroren weefsel van hun eigen testikel stamcellen op te kweken. Bij muizen is het gelukt om deze cellen terug te plaatsen in een testikel. Deze muizen krijgen gezonde nakomelingen. Nu is de tijd aangebroken om de therapie voor mensen te ontwikkelen.

Bij de productie van geneesmiddelen is het essentieel om de juiste stereo-isomeer te isoleren. Andere stereo-isomeren van dezelfde stof zijn in het beste geval minder biologisch actief en soms zelfs schadelijk. Maar de analyse van stereo-isomeren is niet eenvoudig een vaak tijdrovend en arbeidsintensief.

De lichaamseigen, cannabisachtige stof 2-AG stimuleert alcoholgebruik. Onderzoekers vonden een stof die de vorming van dit endocannabinoïde remt. Dat zorgt ervoor dat alcoholverslaafde muizen minder alcohol gebruiken.

Het succes van celtherapie betekent wel dat geschikte cellen op grote schaal en betaalbaar geproduceerd moeten worden. Een Israëlisch biotechbedrijf bedacht een oplossing met placentacellen.

Bioprocestechnologie is van oudsher een batchtechnologie, waarbij cellen in een gesloten bioreactor enkele weken groeien en daarna geoogst worden. Maar sinds een aantal jaar werken fabrikanten aan het continu maken van dit proces. Met name de kolomchromatografie voor zuivering wordt aan gewerkt. Wat is er bereikt en wat zijn de uitdagingen? Experts van vier bedrijven uit het veld geven een inkijkje.

Pijn zit tussen je oren. Letterlijk, want het signaal dat er ergens iets mis is in je lijf wordt in je hersenen vertaald naar ‘ik heb pijn’. En dat is maar goed ook, want pijn zorgt ervoor dat we maatregelen nemen om de oorzaak van de pijn weg te nemen of te verminderen. Maar er zijn verschillende soorten pijn, zoals bij een verwonding of ziekte, of acute en chronische pijn. Pijn is niet voor iedereen hetzelfde. Zo zijn er genetische verschillen en verschillen tussen mannen en vrouwen. Daarom zijn er veel verschillende klassen pijnstillers met ieder hun eigen werking en toepassing. De vrij verkrijgbare pijnstillers zoals paracetamol en ibuprofen kent iedereen. Werken die niet voldoende, dan grijpt de arts naar krachtiger pijnstillers zoals morfine, opioïden genoemd. Die stoffen werken meestal erg goed tegen de pijn, maar hebben grote nadelen zoals bijwerkingen en een kans op verslaving. Daarom zoeken artsen en onderzoekers naar nieuwe methoden om pijn te bestrijden en daarbij kijken ze niet alleen naar nieuwe chemische middelen.

Ouder worden gaat vanzelf, maar veroudering is een gecompliceerd proces. Alles wat bijdraagt aan het verval van een systeem; een mens, dier of ding, is veroudering. Slijtage, ziekte en sterven zijn onderdelen ervan. Mensen en de meeste andere organismen verouderen deels doordat onze genen niet perfect staan afgesteld. Maar we verouderen ook door pech: we staan bloot aan bijvoorbeeld zonnestraling of toxische stoffen waardoor we eerder ouder worden. En gezond leven draagt weer bij aan het vertragen van het proces.

Many IMM graduates pursue a career in academia. But just as many of them venture out into an industrial career to contribute to the valorization of scientific research. Silvie Meeuwissen is one of those scientists. After a successful but fundamental PhD project she preferred the more applied research at ChemConnection, a contract research and manufacturing company. From the state of the art laboratory of the company in Oss she tells us why.

Een muis met een mensenoor op zijn rug baarde eind jaren negentig opzien. Wetenschappers waren erin geslaagd een bioafbreekbare mal in de vorm van een oor te bekleden met cellen en te plaatsen onder de huid van een muis, zodat de cellen gaan groeien. Nu was dit geen echt, werkend oor, maar het principe was bewezen: het is mogelijk om cellen te laten groeien op een drager, zodanig dat ze oud of kapot weefsel kunnen vervangen en vernieuwen.

Many NPC projects are coming to an end. The PhD students involved are about to be released onto the career market. Many of them are not sure about what to do after their graduation, as became clear when the application was opened of the BioBusiness Summerschool 2012. The summerschool introduces the students to 'the business world of Life Sciences'. As many as 70 PhDs attended the week long summerschool in Amsterdam to hear about opportunities in large companies, the possibilities to start a business and to have opportunities to extend their network.

Jan Smeitink's purpose in life is finding a cure for mitochondrial diseases. Every year he sees hundreds of children with genetic mitochondrial defects in his clinic. The majority of the kids do not live to be adults. The energy factories of their cells, the mitochondria, fail to produce enough energy for other cellular processes. The result is a plethora of multisystem diseases. The IOP Genomics project has resulted in the development of a high-throughput screening platform for live cells using state-of-the-art imaging techniques. Hundreds of potential drugs for mitochondrial disorders can be tested using this platform. In the near future, this will lead to a cure for some of the mitochondrial diseases.
 

Predicting biological activity of compounds accurately is of great importance in the development of new drugs. Not only bioinformatics methods are used to predict for example receptor interaction with a drug, but also cheminformatics methods to predict the possible toxicity of a potential drug molecule. Jeroen Kazius constructed extensive databases and designed new data mining methods which can more accurately predict such activities.

Aspergillus niger, well known as the fungus that produces black mold on old bread, forms an extended network of interconnected hyphae. This mycelium has an enormous capacity to secrete proteins and other compounds. Its productivity has led the food and pharmaceutical industry to use A. niger  as the preferred biological production factories for, among others, enzymes. New information about genetic signalling in the fungus cells has urged Marc van der Maarel to study the dynamics of the A. niger mycelium in more detail.

Not all old people are equally old. That is, their chronological age - the number of years they have lived - is not the same as their biological age - their life expectancy and health. Some people appear to have a hereditary taint for longevity. By studying a large group of very old people, who are obviously protected from ageing and disease, researchers are trying to find these genetic variations and the biomarkers by which they are expressed.

Mitochondrial disorders are a group of serious, often fatal diseases. Due to a genetic defect, the powerhouses of our cells are unable to produce enough energy from food. The mechanisms are poorly understood, let alone a therapy being available for patients. Jan Smeitink works to understand the energy metabolism in mitochondria by setting up a screening platform. This platform should generate enough knowledge to influence the energy metabolism and eventually cure disorders associated with it.

The future of personalized medicine ultimately depends on two factors, says Martijn Groot of Novartis. One is the ability of pharmaceutical companies to manage the costs of development and the other is the increasing cost of healthcare. "How much are we willing to pay?"

Jos Beijnen defends the 'provocative statement' that in oncology each patient has always received an individualized  treatment. But new discoveries make even further personalization possible. However, careful selection of patients plays a crucial role in personalized medicine, Beijnen argues.

Om dieren vet te mesten, moeten ze gezond blijven en daarvoor gebruiken we in Nederland maar liefst 590 ton antibiotica per jaar. Het gevolg: resistente bacteriën die ook bij mensen voor problemen kunnen zorgen. Een verbod op antimicrobiële groeibevorderaars in varkensvoer is niet genoeg om het tij te keren.

De NVFG is binnen de internationale organisatie voor farmaceutische geneeskunde IFAPP een van de grootste verenigingen. Toch geniet het vak van farmaceutisch arts weinig bekendheid. Rudolf van Olden, farmaceutisch arts en medisch directeur van GSK, licht toe.

Tweeduizendzeven was het jaar van de start van de projecten voor het Top Instituut Pharma. Het topinstituut is in juli 2006 officieel gelanceerd. Het is uniek als zogenaamd public-private partnership waarin academia, bedrijfsleven en regulatoire instellingen samenwerken. Het doel is de efficiëntere ontwikkeling van geneesmiddelen waardoor de patiënt sneller en goedkoper genezen kan worden. Met het uitvoeren van een sterk R&D programma, het opzetten van een trainingsprogramma en het zoeken van de samenwerking buiten de grenzen werkt het topinstituut aan het vestigen van zijn naam.

Monoclonal antibodies are the quickly rising stars of cancer therapeutics. One of the first fully human therapeutic antibodies is developed by the partly Dutch biotech company Genmab. The first trials with this therapeutic, HuMax-CD20, show amazing results against B-cell chronic lymphocytic leukemia. Genmabs EVP and CSO Jan van de Winkel is thrilled about the success. "I think this is one of the best examples of an academic concept taken to the clinic."

A drug delivery system based on a dextran hydrogel is on the virge of commercialization. Prof.  Wim Hennink incorporates therapeutic proteins into dextran-based microspheres. These proteins are released by diffusion and degradation of the hydrogel. Timing of release is crucial and can be precisely tuned. Hennink has been working on this new system since the early nineties. The pharmaceutical company Octoplus is now 'valorizing' it by taking it to the clinic. "Valorisation is the translation step needed to go from academic concept to industrial product."

GlaxoSmithKline koopt de rechten op HuMax-CD20 van Genmab, een geneesmiddel op basis van menselijke antilichamen tegen onder andere chronische lymfatische leukemie. De totale overeenkomst behelst ruim twee miljard dollar. Dat is de grootste deal ooit gesloten in de biotechnologie-sector.

Vorige maand ondertekenden drie partijen de tweede onderzoeksovereenkomst binnen het Top Institute Pharma. De Universiteit Leiden/LACDR, het UMC St. Radboud Nijmegen en het Deense farmaceutische bedrijf Lundbeck sloten een overeenkomst voor de uitvoering van een vierjarig fundamenteel onderzoek naar schizofrenie.

Twee jaar geleden richtten klinisch chemici Hans van Duijnhoven en Christian Schoenmakers de Stichting ServiceLabs op. Het is de eerste organisatie die zich richt op professioneel uitgevoerde zelfdiagnostiek voor particulieren. Hoe gaat het nu met de ondernemers en de stichting?

De KNAW vindt dat de overheid moet investeren in het koppelen van databanken met gegevens en lichaamsmateriaal van patiënten, zogenaamde biobanken. Dan kan beter onderzoek worden gedaan naar multifactoriële aandoeningen.

Dr. Maytal Bivas-Benita ontwikkelde nieuwe toedieningsvormen voor een DNA vaccin en bewees dat deze beter werken wanneer ze direct in de long worden toegediend. Daarvoor ontving zij tijdens de FIGON Geneesmiddeldagen de NVFW-Octoplus Thesis Award.

"Two-thirds of all medicine has a background in food. Research at the interface between food and pharma is important," says Dr. Johan Garssen. In a strategic alliance between Numico Research and the UIPS in Utrecht he investigates the influence of certain oligosaccharides in breast milk on the immune system. These sugars act as prebiotics and in one trial reduced the incidence of allergy in babies by half. "This group of oligosaccharides is capable of much more than we ever imagined."
